We administer and dispense drugs to our patients every day. Yet how much do we really understand about the factors that will influence the drug effect? Why can one patient have no side effects for a commonly used drug, yet another might suffer a life-threatening reaction? In this lecture, we will review the principles underlying drug pharmacodynamics (what the drug does to the body) and pharmacokinetics (what the body does to the drug) and the factors that can affect them. Using examples of common drugs used in veterinary practice, we will consider key questions such as: What is the impact of different routes of administration on drug effect? What factors will limit drug access to certain tissues and when does it matter? Do all drugs need to be metabolised? What patient factors will alter absorption, distribution, metabolism and elimination and thus increase the risk of an adverse drug reaction or reduced efficacy?
